Bacterial community structure and predicted functional pathways in sugarcane rhizosphere soils under legume intercropping
Abstract
Legume intercropping is a sustainable agronomic strategy that benefits sugarcane crops. This study characterized shifts in bacterial community structure, carbon-source oxidation, and predicted metabolic pathways in sugarcane rhizosphere soils under cowpea, mung bean and peanut intercropping. Rhizosphere samples were analyzed via 16S rRNA gene amplicon sequencing (V3-V4), substrate oxidation and functional prediction using the PICRUSt2 pipeline. Intercropping enhanced the metabolic activity and diversity measured by in vitro community-level physiological profiling, which showed an increase in carbohydrate and carboxylic acid consumption. Although taxonomic alpha and beta diversity showed no global differences, differential abundance analysis showed changes in specific biomarkers. Legume intercropping increased the abundance of Actinobacteriota, specifically enriching Acidimicrobiales and Actinomycetia, with genera such as Actinoplanes and Nocardioides identified as key indicators. Co-occurrence network analysis indicated a modular structure comprising specialized ecological niches, with keystone taxa including members of Actinobacteriota, Chloroflexota, and Nitrososphaeraceae. Predicted functional profiles were dominated by carbohydrate and amino acid metabolism, with intercropping enriching pathways for xenobiotic degradation and the biosynthesis of siderophores and arabinogalactan. Intercropping was also associated with predicted enrichment of genes related to glutathione and amino acid metabolism. In contrast, sugarcane monocultures showed predicted biomarkers potentially associated with nutritional stress. These results suggest that legume intercropping influences specific microbial groups and predicted metabolic pathways related to amino acid degradation and biosynthesis, indicating potential benefits for microbial activity and nutrient cycling.
